Lot was Abraham’s nephew but he is called brother Gen 13:8 Abraham and lot bothers Mt 13:55 Is not this the carpenter's son? is not his mother called Mary? and his brethren, James, and Joses, and Simon, and Judas? 56 And his sisters, are they not all with us? Whence then hath this man all these things? Galatians 1:19 But other of the apostles saw I none, save James the Lord's brother. not biological brothers and sisters, not children or Mary the Mother of God for sure! but if Jesus has brothers why is only James called the “brother of the Lord”? You do realise the purpose of the commas is to identify separate people, as follows: (1) Mary, the mother of Jesus; (2) Mary's sister, Salome, the mother of John; (3) Mary, the wife of Cleopas; (4) Mary Magdalene.
