According to my understanding, speaking against the Holy Spirit would mean that a person who is absolutely sure of the Truth of God's Word, would wilfully reject and deny Him. In reference to the scripture you posted it was about people who speak against the Holy Spirit, not others. This was in reference when the Pharisees and Scribes were calling Jesus miracles he performed are from Beelzebub the prince of demons. They were calling the Holy Spirit a demonic spirit. But when the Pharisees heard it, they said, "It is only by Beelzebul, the prince of demons, that this man casts out demons." ... And if I cast out demons by Beelzebul, by whom do your sons cast them out? Therefore they will be your judges. But if it is by the Spirit of God that I cast out demons, then the kingdom of God has come upon you. ... Therefore I tell you, every sin and blasphemy will be forgiven people, but the blasphemy against the Spirit will not be forgiven. And whoever speaks a word against the Son of Man will be forgiven, but whoever speaks against the Holy Spirit will not be forgiven, either in this age or in the age to come. - Mat 12:24, 27-28, 31-32 I think this is a very scary warning that we all need to listen to very carefully. Yes, your scripture is talking about speaking against the Holy Spirit. As has been said, ones were saying it was not God working miracles through Jesus. Like this, I would say, you can speak against people by making up things to say about them, when you do not have proof those things are true.
