Could be it is wet shriveled up toilet paper! If the drain goes to the house sewer system or septic tank and is the lowest and/or closest open drain to the main, or is not vented within five feet. You may have even suffered a back up from the street sewer into your hot tub.

Locally there is a huge area of the city that has that problem. Old city storm sewers and black water sewer pipes are failing, leaking one into the other. When we get heavier rains they  overflow and back up into houses. They are experimenting with four inch flapper valves for houses here. The flapper valve allows  bad stuff out but snaps closed if bad stuff flows the other way. Anyway, you could have had stuff come in and then drain out leaving a nasty residue. Had it happen to an entire school building- Oh my  we had to call in the professionals to sanitize after that one.

Also some mighty nasty stuff can gather for a long time in the filter for the tub's recirculation pump. If you used heavy cleanser/dissolver perhaps it did it's job and dissolved what was caught in the trap allowing it to float back and say hello. If there is a trap or filter did you empty it?
